Development of a New Polymeric Nanocarrier Dedicated to Controlled Clozapine Delivery at the Dopamine D2-Serotonin 5-HT1A Heteromers

Clozapine, the second generation antipsychotic drug, is one of prominent compounds used for treatment schizophrenia. Unfortunately, use this drug still limited due to serious side effects connected its unspecific and non-selective action. Nevertheless, clozapine remains first-choice situation drug-resistance Development new strategy delivery into well-defined parts brain has been a great challenge modern science. In present paper we focus on presentation nanocarrier targeted transport, enabling interaction with dopamine D2 serotonin 5-HT1A heteromers (D2-5-HT1A) in tissue. Clozapine polymeric nanocapsules (CLO-NCs) were prepared using anionic surfactant AOT (sodium docusate) as an emulsifier, bio-compatible polyelectrolytes such as: poly-l-glutamic acid (PGA) poly-l-lysine (PLL). Outer layer carrier was grafted by polyethylene glycol (PEG). Several variants nanocarriers containing varying physicochemical parameters tested. This kind approach may enable availability safety improve selectivity action, finally increase effectiveness schizophrenia therapy. Moreover, purpose manuscript cover wide scope issues, which should be considered while designing novel means delivery. It important determine interactions many cell components various cellular levels order sure that will safe won’t cause undesired patient.
